A fusion gene is a chimeric gene formed by the fusion of partial sequences of two genes, usually due to chromosomal translocations or deletions. These chimeric genes can form abnormal transcripts or proteins in subsequent biological processes, which can lead to or promote the development of tumors. The fusion transcripts are not necessarily all derived from fusion genes. In contrast, sequencing-based methods such as whole genome sequencing (WGS) and RNA sequencing are widely used to identify previously unidentified gene fusions. Several bioinformatics methods have been developed to detect fusion transcripts from RNA-Seq data, such as ChimeraScan, SnowShoes-FTD, TopHat-Fusion, FusionMap and FusionSeq.

coli are often used for the production of small and simple enzymes due to their ease of use and low cost. However, the expression of more complex proteins or proteins requiring post-translational modifications such as glycosylation or phosphorylation may require more advanced expression systems such as yeast, insect or mammalian cells. coli expression systems, and the T7 promoter system, which is often used in bacterial and insect cell expression systems. Recombinant DNA technology is often used to produce enzymes, while chromatography is the most widely used method for enzyme purification. Advanced purification techniques such as electrophoresis, ultrafiltration, and crystallization may be used to further purify the enzyme.

In recent years, enzymes have been widely used in disease diagnosis, providing a quick and accurate way to detect and monitor diseases. These enzymes are often elevated in the blood of patients with infectious or inflammatory diseases and are used as indicators of disease severity. Liver enzymes such as alanine aminotransferase (ALT) and aspartate aminotransferase (AST) are used to diagnose liver disease. Elevated levels of these enzymes indicate liver damage or dysfunction, and monitoring their activity can provide valuable information for disease management. As research continues to advance, we can expect to see even more exciting applications for enzymes in disease diagnosis in the future.

With the development of high-resolution mass spectrometry (HMS) and its instruments, the qualitative and quantitative analysis of compound quality determination has received more and more attention and been widely used by researchers. The accurate mass measurement method can determine the accurate quality of a compound ion, perform qualitative and quantitative analyses of protein composition, perform quantitative analyses of amino acid structure, examine the composition of metal compounds, and can also be used for the qualitative detection of the majority of small molecule drugs. The difference between the accurate mass of a molecule ion and the accurate mass of a fragment ion can be used to determine the accurate quality. At the same time, some qualitative errors are avoided, and the analysis results are more accurate and reliable. Ft-ICR MS was used to detect and analyze the extract of this traditional Chinese medicine, and the obtained excimer ion peak and secondary mass spectrometry fragment information were compared with the information reported in the literature, and 42 compounds were identified.

The difference in size and strength of each component causes the difference in the residence time of the component to flow out of the stationary phase. Chromatographic separation methods include adsorption chromatography, bonded-phase chromatography, ion exchange chromatography, affinity chromatography, and size exclusion chromatography. It is a liquid chromatography method that uses the binding characteristics of stationary phases to adsorb target products and achieve separation and purification. can be used to form one of the interacting substances with the insoluble support, which can be used as a stationary phase for chromatography. SDS polyacrylamide gel electrophoresis (SDS-PAGE) has the advantages of simple operation and good reproducibility in the determination of protein molecular weight, detection of specific proteins, and identification of strain species. Positively correlated with the molecular weight of the protein, the length of the complex formed by proteins of different molecular weights is different. SDS makes the amount of negatively charged protein far exceed its original charge, masking the natural charge difference between various protein molecules. Therefore, the SDS-treated protein molecules move between the upper glycine molecule and the lower Cl ion. In the laboratory, a standard molecular weight protein covalently coupled to a dye is used as a reference protein to roughly indicate the size of the unknown protein.

Introduction to Genome-wide Association StudiesA genome-wide association study is what the National Institutes of Health call "a study of common genetic variation across the whole human genome with the goal of finding genetic links to observable traits. The majority of common known SNPs are genotyped in each group's members. The exact number of SNPs varies depending on genotyping technology, but it is usually one million or more. Bioinformatics software such as SNPTEST and PLINK, which support many of these alternative statistics, is commonly used for calculations. Identifying statistically relevant interactions in GWAS data is computationally and statistically difficult due to the potentially infinite number of interactions.

In addition to this basic function, RNA-seq can be used for differential gene expression, variant detection and allele-specific expression, small RNA profiling, characterization of alternative splicing patterns, system biology, and single-cell RNA-seq. Most initial small RNA discovery studies used pyrosequencing and, subsequently, other NGS platforms with higher throughput, which resulted in genome-wide surveys and the discovery of an increasing number of small RNA species. Paired-end sequencing enables sequence information from both ends, thereby detecting splicing patterns without a requirement for previous knowledge of transcript annotations. In addition, a majority of living cells cannot be cultivated in vitro, single-cell RNA-seq may discover novel species or regulatory processes of biotechnological or medical relevance. The workflow of single-cell RNA-seq generally involves the following steps: single-cell isolation, cDNA library construction, RNA-seq, and bioinformatics.
