Exploration of Digestion Method for Determination of Heavy Metal Elements in Soil by ICP-MS

Air pollution and pesticide residues will continue to accumulate in the soil. Mercury is ubiquitous in nature. If the medicinal materials grown in mercury-polluted soil are eaten by the human body, the content of heavy metal elements such as metallic mercury, inorganic mercury, and organic mercury will accumulate until the amount that the human body can bear, therefore, more and more attention has been paid to the detection of mercury and heavy metals in medicinal soil.

Mercury is a volatile heavy metal element, which cannot be accurately quantified by the heating and digestion method in the commonly used detection. The micro-source testing laboratory consulted literature and shared a method of using microwave digestion ICP-MS to detect medicinal material cultivation soil.

Use 1% nitric acid to make 1000μg·L-1 mercury standard solution to make a standard curve with a concentration of 0.1, 0.5, 5, 10, 15μg·L-1, accurately weigh 0.05g soil sample after passing through a 100-mesh sieve, put it in a microwave digestion tank, add a certain proportion of concentrated nitric acid, hydrofluoric acid and perchloric acid, cover it tightly, and put it in a microwave digestion instrument. After digestion, put it on the electric heating plate, add water to catch the acid twice, until about 1 drop remains, transfer it to a 50mL volumetric flask with ultrapure water, and set the volume to the machine.

Draw 1mL of mercury standard solutions with different concentrations, and measure the count value in the ICP-MS instrument. The results showed that the correlation coefficient of the mercury standard curve was 0.9994. Draw 1mL of the sample digestion solution and inject 6 times continuously. The intraday RSDs of mercury were 2.94% and 2.59%, and the intraday RSDs were 2.32%. It can be seen that the precision of this method is good.

The mercury standard solution was added to 6 soil samples with known heavy metal content, and the samples were made into digestion solution, and the average recovery rate of mercury was 94.07%, which showed that the detection method was highly accurate.

The microwave digestion method can not only heat quickly, but also meet the high pressure digestion under low temperature conditions. Compared with the conventional heating digestion method, it has the advantages of complete digestion, low blank value and high accuracy.

The ICP-MS detection method used has a good linear relationship and has the advantages of high accuracy, good precision, simplicity and speed. In addition to mercury heavy metal elements, the content of other heavy metal elements in preparations or APIs can also be analyzed by ICP-MS.
